Platelets represent a decisive factor in (cardio-)vascular diseases, the leading causes of death in the industrialised world. In blood vessels platelets are preserved in a sensitive balance between upholding adequate blood-flow and promoting blood clotting in the case of bleeding. Upon vessel injury, platelets instantly become activated, leading to granule release, aggregation, and plug formation. Although much is known about factors resulting in platelet activation, the molecular characteristics of platelets maintained in their balanced state are less extensively characterized. Since an enhanced platelet reactivity represents an important factor for thrombotic complications, it is necessary to explore biomarkers ensuring platelets' activation status for their monitoring in health and disease.

The aim of this work will be to gain a better understanding how platelets contribute to an increased risk of thrombosis. Thus, the platelet proteome of patients with increased risk of thrombosis, healthy controls and in vitro activated as well as in vitro inhibited platelets should be investigated. These investigations include the analysis and comparison of the platelet proteome of all these conditions by the proteomics technology of fluorescence two-dimensional differential in gel electrophoresis (2D DIGE).

Qualifications required:

We are looking for a master student highly interested in clinical research for our group at the Center of Physiology & Pharmacology (Medical University Vienna) and Clinical Division of Hematology and Hemostaseology, Department of Medicine I and. We expect you to have a theoretical knowledge in molecular biology, teamwork skills, motivation to conduct a challenging master thesis, interested in translational science, dependability and ability to work independently.

Opportunities and methods:

Students will have the opportunity to learn the isolation techniques of platelets and a range of biochemical analysis methods: Platelet proteomics via 2D-DIGE, protein identification, validation via mass spectrometry and Western blot as well as platelet functional analysis by flow cytometry.

Starting date: starting time as soon as possible, minimal duration 10-12months
