The Laboratory of Single-Molecule Genetics at the Department of Biophysics, Johannes Kepler University (JKU) Linz, is inviting applications for a PhD position in Molecular Biology starting September 2025. We are seeking a motivated and independent candidate with a strong interest in applying cutting-edge technologies to address fundamental questions in genome biology.

Project Description

The doctoral research project will investigate repeat instability in the genome, with a focus on meiotic recombination hotspots—genomic regions prone to elevated mutation rates during meiosis. The project involves:

Construction of repeat cassettes, CRISPR-based gene editing, High-throughput and error-corrected next-generation sequencing (NGS) [1,2], PCR-based assays, Advanced data analysis using computational tools and artificial intelligence [3,4]

This work is embedded in a larger research consortium comprising nine research groups from the Vienna BioCenter, IST Austria (ISTA), and JKU, applying complementary state-of-the-art methodologies to elucidate mechanisms of genome evolution during meiosis.

The aim is to elucidate whether unique sequence features in recombination hotspots contribute to elevated mutagenesis and how these patterns differ from other genomic regions or mitotic divisions. The insights gained may inform our understanding of mutation-driven evolutionary processes and genetic disease mechanisms.

Candidate Profile

We welcome applications from early-stage researchers with a strong background in molecular biology and genetics, interest or experience in evolutionary biology, population genetics, and/or bioinformatics (preferred but not required), proficiency in English (the working language of the lab), experience with NGS, molecular cloning, or computational analysis of genomic data is advantageous.

Funding and Duration

The position is funded for 3.5 years, starting September 2025 (negotiable), and will be paid according to the Austrian Science Fund (FWF) salary scale.

Application Instructions

Applicants should submit a single PDF file including:

1. A cover letter outlining your motivation, research interests, and suitability for the position

2. A Curriculum Vitae, including a list of technical skills and any publications

3. Contact details for 2–3 referees who are willing to provide reference letters

Please send your application to: irene.tiemann(at)jku.at

Review of applications will begin immediately and continue until the position is filled.
