Die Organe unseres Körpers werden durch so genannte Stammzellen am Leben erhalten. Stammzellen sind einzigartig, weil sie sich selbst erneuern können. Diese Eigenschaft erlaubt es ihnen, identische Kopien von sich selbst zu erzeugen, aber gleichzeitig auch andere Zellen zu erzeugen, die sich spezialisieren und wichtige Aufgaben in den verschiedenen Organen übernehmen. Dadurch bleiben die Stammzellen selbst über lange Zeit erhalten während sie ständig absterbende oder beschädigte Zellen ersetzen. In unserem Gehirn und Rückenmark jedoch ist die Situation jedoch etwas anders. Hier erfüllen sehr viele verschiedene Arten hoch spezialisierter Nervenzellen ganz individuelle Aufgaben. Einige aktivieren und andere hemmen neuronale Schaltkreise. Woher wissen die Stammzellen, welche Arten von Nervenzellen sie an welchen Orten und zu welchem Zeitpunkt erzeugen sollen? Wie verändern sich die Stammzellen während der Zeit, so dass sie die richtigen neuronalen Subtypen erzeugen können? Ziel unseres SFB ist es, die Mechanismen zu verstehen, die neurale Stammzellen in Raum und Zeit modulieren. Wir wollen herausfinden, wie neuronale Stammzellen es schaffen, sich einerseits selbst zu erhalten, andererseits aber zu verändern, um grundlegend verschiedene Arten von Nervenzellen zu erzeugen. Um dieses Problem anzugehen, kombiniert unser Konsortium zahlreiche methodische Ansätze, die um einige zentrale Schlüsseltechnologien gruppiert sind. Wir setzen eine Reihe verschiedener Modelle ein - von Meereswürmern bis hin zu menschlichen 3D-Zellkulturen. Wir konzentrieren uns auf Einzelzelltechnologien, die es uns ermöglichen, Stammzellen in Raum und Zeit genau zu verfolgen und für jede Zelle eine Liste aller aktiven Gene zu erzeugen. Im Mittelpunkt unseres Konsortiums steht eine zentrale Bioinformatik Einheit, die es uns erlaubt, die großen dabei entstehenden Datensätze zu interpretieren und in Verbindung zu setzen. Gemeinsam wollen wir einen zentralen Drehpunkt für die neurale Stammzellbiologie in Österreich aufbauen, der eine einzigartige, vereinheitlichende biologische Frage mit Hilfe modernster Technologien an der Spitze des technologisch Möglichen angeht.
