Zelluläre und biologische Prozessewerdenunter anderem durch Änderungen von Ionenkonzentrationen innerhalb sowie außerhalb der Zelle bestimmt. Ionen werden über spezielle Poren, den sogenannten Ionenkanälen, über die Zellmembran transportiert. Dieses Projekt trägt dazu bei, das Zusammenspiel von Kalzium (Ca2+) und Kalium (K+) durchlässigen Ionenkanälen und deren Rollen in der Entstehung von Prostata- und Darmkrebs zu verstehen. Ein besonders wichtiger Eintrittsweg für Kalzium in die Zellen ist der sogenannte Ca2+ release- activated Ca2+ (CRAC)-Ionenkanal. Tritt Kalzium durch diesen in die Zelle ein, werden eine Vielfalt von Proteinen wie auch Ca2+ aktivierte K+ Ionenkanäle reguliert. Defekte in diesen Ionenporen oder ein verändertes Vorkommen dieser Ionenkanäle in der Zelle kann unter anderem auch zur Entstehung von Krebs führen. Somit sind diese Ionenporen von hoher klinischer Relevanz. Im Besonderen wurde gezeigt, dass eine Ko-Regulation von diesen unterschiedlichen Arten von Ionenporen die Entstehung und das Wachstum von Brust-, Darm- und Prostatakrebs steuern kann. Die detaillierten molekularen Bedingungen für die Wechselwirkung dieser Ionenkanäle sind jedoch derzeit unbekannt und sollen daher im Rahmen dieses Projektes identifiziert werden. Dafür werden funktionelle, fluoreszenzmikroskopische sowie biochemische Analysen durchgeführt, um essentielle Bereiche in diesen Ionenporen zu identifizieren, die deren Zusammenspiel bestimmen. Weiterführend wird getestet, ob eine Störung solcher Schlüsselregionen, die Ko-Regulation der jeweiligen Ionenkanäle beeinträchtigt und folglich das Krebswachstum hemmen kann. Zusammenfassend werden unsere Studien dazu betragen ein besseres Verständnis für jene molekularen Faktoren zu erlangen, welche zur Prostata- und Darmkrebsentstehung beitragen, und bieten eine Basis für die Entwicklung neuer therapeutischer Ansätze.
