Abstract

Das Plattenepithelkarzinom und das Basaliom zählen zu den häufigsten humanen Tumoren. Trotz der Datenflut an biologischen Erkenntnissen ist das Verständnis ihrer Entstehung und Entwicklung weiterhin unklar. Es gibt Modelle um die Tumorigenese zu studieren in denen mittels definierter Onkogene Zellen transformiert und zu einer unkontrollierten Proliferation bzw. invasivem Wachstum gebracht werden. Merkmale der malignen Koversion und Metastasierung sind die Herabregulierung von bestimmten Zelladhäsionsmolekülen und die Überexpression von Proteinen der dermoepidermalen Junktionszone wie z.b. Laminin 5, BP180, a6ß4 Integrin und Kollagen VII. Das kürzlich von uns etablierte "Ras Driven"-Tumormodell erlaubt uns die produktion von Plattenepithelkarzinomen. Un tersuchungen am "Ras Driven"-Tumormodell zeigten dass a6ß4 Integrin und Laminin 5 notwendig sind fuer die Entstehung des Plattenepithelkarzinoms. In dieser Studie werden wir die Rolle von BP180 und Kollagen VII in diesem Tumormodell studieren. Ras und Iba werden in BP180- bzw. Collagen VII- defizienten Keratinozyten koexprimiert und dann subkutan in Mäuse injiziert. Als Kontrolle werden Keratinozyten von gesunden Patienten benutzt. Um zu überprüfen ob die Tumorprogression zu verhindern wäre, werden wir in existierenden Tumoren BP180 und Kollagen VII blockierende Antikörper injizieren. Falls eins oder beide der Proteine notwendig ist für Tumorigenese werden wir die Bedeutung der verschiedenen Protein-Domänen studieren. Unser Studienziel ist festzustellen welche Rolle hemidesmosomale Proteine im Tumor-Entstehungsprozess spielen.
