Microsatellites are one of the most commonly used genetic markers. Despite this, their mutation processes are still poorly understood. In particular, the large variation in microsatellite density and distribution among species remains an open question. This proposal focuses on the mechanistic basis of microsatellite instability and its possible implications for genome evolution. After introducing heterologous msh-2 genes in C. elegans microsatellite mutation rates and pattern will be compared for transgenic C. elegans carrying msh-2 genes from different species. Most important, the observed pattern will be compared against the expectation based on the genomic distribution of microsatellites in the species from which the orthologous msh-2 genes were isolated. A screen for new genes affecting microsatellite stability in E. coli will complement this detailed characterization of one gene influencing microsatellite stability.
