Unsere Gruppe ist daran interessiert, einerseits die normale Entwicklung des menschlichen Gehirns zu erforschen und andererseits zu verstehen, wie sich diese durch Krankheiten verändert. Eine große Herausforderung bei der Erforschung der Biologie und Krankheit von menschlichem Gewebe ist die Zugänglichkeit des Materials. Um dieses Hindernis zu überwinden wurde im Knoblich Labor eine neuartige bahnbrechende Technologie, die sogenannten Hirnorganoide entwickelt. Dieses 3 D Modellsystem wird aus menschlichen Stammzellen gewonnen und kann humanspezifische Aspekte der Gehirnentwicklung rekapitulieren. Die Verwendung von Organoiden ermöglicht uns die Modellierung von Erkrankungen, die das Nervensystem betreffen, und bietet somit das Potenzial, die Krankheitsforschung grundlegend zu verändern. In diesem Projekt wollen wir die Technologie der Hirnorganoide weiter vorantreiben, indem wir neurologische Krankheiten im Labor nachbilden, um diese fundamental verstehen zu können und letztlich die Entwicklung neuartiger Therapien zu ermöglichen. Wir stellen die Hypothese auf, dass der Austausch zwischen Zellen verschiedener Hirnregionen in unserem Modellsystem untersucht werden kann, was es zu einem wertvollen Instrument macht, um die Anfangsstadien einer bestimmten Hirnstörung zu untersuchen, bevor diese zu einer erkennbaren Pathologie wird. Um dies zu erreichen, werden wir Organoide erzeugen, die verschiedene Hirnregionen, die mit bestimmten Hirnkreisläufen verbunden sind, auf eine für die Entwicklung repräsentative Weise zusammenführen. Dadurch können wir zum ersten Mal neurologische Interaktionen zwischen diesen Regionen in einer Petrischale untersuchen. Dieses neue Organoidmodell wird es uns ermöglichen, krankheitsbezogene Aspekte eines neurologischen Schaltkreises zu untersuchen, auf den Suchtmittel und pharmazeutische Wirkstoffe besonders abzielen. Das vorgeschlagene Projekt wird die Pathologie neurologischer Erkrankungen im Labor nachbilden und damit weit über die bestehenden Modelle hinausgehen und ein innovatives menschliches Krankheitsmodell schaffen, das die bestehende Forschung zu diesen neurologischen Störungen verfeinern und ergänzen kann.
Übersetzter Abstract Während Tiermodelle in der Vergangenheit grundlegende Erkenntnisse für die Neurowissenschaft geliefert haben, erfordert die Entwicklung präziser Behandlungen für menschliche neurologische Erkrankungen die Etablierung menschenspezifischer Modelle, um deren Einsatz zu reduzieren und letztendlich zu ersetzen. Das menschliche Gehirn ist auf einen chemischen Botenstoff namens Dopamin angewiesen, um unsere Bewegungen zu steuern und Belohnungsgefühle zu verarbeiten. Wenn dieses System gestört ist, kann dies zu schweren Bewegungsstörungen wie der Parkinson-Krankheit führen oder zu psychiatrischen Erkrankungen und Sucht beitragen. Da sich tierische Gehirne anders und viel schneller entwickeln als menschliche Gehirne, können bestimmte Aspekte menschlicher Krankheiten - wie der spezifische Verlust von Dopamin bei Parkinson - in Tieren nicht vollständig oder genau nachgebildet werden. Um diese Lücke zu schließen, hat unser Forschungsteam erfolgreich lebende, dreidimensionale Modelle des Belohnungs- und Bewegungssystems des menschlichen Gehirns entwickelt, um neurologische Erkrankungen ohne Rückgriff auf Tierversuche zu untersuchen. Wir verwendeten Stammzellen, um miniaturisiertes menschliches Gehirngewebe, sogenannte "Organoide", zu züchten, wobei wir uns speziell auf drei verschiedene Regionen konzentrierten: das Mittelhirn (in dem Dopamin produziert wird), das Striatum und den Kortex. Indem wir diese Gewebe in einer maßgeschneiderten Form nebeneinander platzierten, verschmolzen sie auf natürliche Weise zu einer einzigen, verbundenen Struktur. Bemerkenswerterweise bildeten die Dopamin produzierenden Zellen im Mittelhirn lange, funktionelle Verbindungen, die sich in die anderen Gehirnregionen erstreckten und diese vernetzten, exakt so, wie es in einem sich entwickelnden menschlichen Gehirn der Fall ist. Wir konnten bestätigen, dass diese im Labor gezüchteten Netzwerke elektrisch aktiv und vollständig in der Lage waren, Dopamin freizusetzen. Dieser technologische Sprung hat tiefgreifende Auswirkungen auf die Medizin, die Gesellschaft und die Forschungsethik. Erstens treibt er die Entwicklung von Parkinson-Behandlungen voran. Indem wir gesunde, Dopamin produzierende Stammzellen in unser Modell injizierten, konnten wir beobachten, wie sie überlebten, reiften und sich in das bestehende Gehirnnetzwerk integrierten. Dies belegt, dass dieses System ein hervorragendes Testumfeld für vielversprechende Stammzell-Ersatztherapien darstellt. Zweitens bietet es beispiellose Einblicke in die Suchtforschung. Als wir diese Gehirnmodelle Kokain aussetzten, entdeckten wir, dass chronische Exposition zu langanhaltenden Veränderungen der physischen Verbindungen und der elektrischen Aktivität des Gehirns führte, die auch nach Absetzen der Droge bestehen blieben. Am wichtigsten ist jedoch, dass dieses menschenspezifische Modell eine leistungsstarke, ethische Alternative zu Tierversuchen bietet. Es unterstützt direkt die "3R"-Initiative (Replacement, Reduction und Refinement / Vermeidung, Verringerung und Verbesserung), indem es eine präzisere und hochwirksame Plattform zum Testen pharmazeutischer Medikamente und zur Erforschung von Gehirnerkrankungen im menschlichen Kontext bereitstellt.
