Das maligne Pleuramesotheliom ist eine seltene Krebserkrankung mit schlechter Prognose. Seit langem ist bekannt, dass Angiogenese (die Bildung von neuen Blutgefäßen) eine wichtige Rolle im Bereich des malignen Tumorwachstums spielt. Neue Medikamente, welche das Ziel haben, sich gegen die Blutversorgung von Tumoren zu richten (Angiogenese-Inhibitoren), sind heute gut in der onkologischen Therapie etabliert. Trotz guter klinischer Resultate dieser Substanzen, bleibt die erwünschte Wirkung jedoch gelegentlich aus (Therapieresistenz). Eine solche Resistenz gegen Angiogenese-Inhibitoren entsteht durch eine Vielfalt von Mechanismen. Eine wesentliche Rolle spielt dabei jedoch das Tumorwachstum ohne entsprechende Angiogenese, wobei der Tumor hier keine neuen Blutgefäße bildet, sondern bereits bestehende Blutgefäße für sich einnimmt (Gefäß-Inkorporation). Weiters wird das Bindegewebe innerhalb von Tumoren für eine Therapieresistenz verantwortlich gemacht: funktionslose Blutgefäße und inhomogene Durchblutung führen zu einer verminderten Verteilung von Angiogenese-Inhibitoren und Chemotherapie innerhalb des Tumorgewebes. Somit werden Tumorzellen, welche nicht unmittelbar neben funktionierenden Blutgefäßen liegen, von nur unzureichenden Medikamentendosen erreicht, was zur Therapieresistenz führt. Das vorliegende Studienprotokoll soll dabei helfen, diese Resistenzmechanismen besser zu verstehen und zukünftig effektivere Therapien für Patienten mit Krebserkrankungen, wie das maligne Pleuramesotheliom, entwickeln zu können.
