Kinasen agieren als zentrale zelluläre Signalmoleküle. Dadurch sind sie aber auch das Ziel von Viren, um in der Folge die infizierte Wirtszelle auf Virenproduktion umzuprogrammieren. Auch der SARS-CoV-2 Virus verfolgt diese Strategie. Wir vertreten die Hypothese, dass die Modulation von zellulären Kinase-Aktivitäten pathologische SARS-CoV-2 Funktionen eindämmen könnte. Die pharmakologische Blockade von Kinasen könnte eventuell neben der Reduktion der Virusproduktion auch die pathologischen Auswirkungen auf die Zellphysiologie verringern. Das Ziel unseres Projektes ist es zwei Kinase-kaskaden, die mit viralen Proteinen wechselwirken, in menschlichen Lungenzelllinien zu charakterisieren. Zunächst verwenden wir einen zellbasierten Kinase-Konformations-Reporter (KinCon),um die AuswirkungvonSARS-CoV-2 Proteininteraktionen auf die Enzymaktivität ausgewählter Kinasen zu bestimmen. Zweitens, analysieren wir den Effekt von bioaktiven chemischen Molekülen auf zelluläre SARS-CoV-2- Funktionen. Drittens, modulieren wir pharmakologisch die cAMP-PKA und MAPK-Kinase- signalwege und bestimmen Veränderungen der Virusassemblierung. Impfstoffe zur Bekämpfung von SARS-CoV-2-Infektionen retten Tausende von Menschenleben. Mutationen können die Wirksamkeit der Spikeprotein-spezifischen Antikörper beeinträchtigen! Wir hoffen, dass die pharmakologische Veränderung von Kinaseaktivitäten eine Strategie für mutations-entkoppelte COVID-19 Therapieansätze darstellen könnte.
In diesem AKUT-Projekt untersuchten wir anhand verschiedener humaner Zelllinien wie eng humane Proteinfunktionen und die SARS-CoV-2-Biologie miteinander verknüpft sind. Das Projekt wurde in Kooperation zwischen der Medizinischen Universität Innsbruck (MUI) und der Universität Innsbruck (UIBK) durchgeführt und fokussierte darauf, wie kleine Moleküle die Enzymaktivität modulieren und dadurch direkt oder indirekt die virale Replikation sowie weitere zelluläre Prozesse beeinflussen. Aufbauend auf unserer früheren Arbeit zu Kinasefunktionen und Mutationen gewannen wir neue Einblicke, wie kleine Moleküle oder Wirkstoffe über unerwartete ON- und OFF-Target Effekte Wirtszellfunktionen verändern. Ein erstes Ergebnis des Projekts war die Identifikation von zwei bekannten Wirkstoffen, die Enzymfunktionen verändern und den SARS-CoV-2-Replikationszyklus beeinflussen. Beim ersten Treffer, Kinetin, deuteten frühere Befunde auf eine Wirkung über Kinasebezogene Funktionen einschließlich des PINK/Parkin-Mitophagiewegs hin, den wir daraufhin vertieft analysierten. Spätere Ergebnisse einer anderen Forschungsgruppe zeigten jedoch, dass die antivirale Wirkung wahrscheinlich unabhängig von der Kinaseaktivierung ist und stattdessen mit RNA-Polymerase-aktivität zusammenhängen könnte. Dieser Befund lenkte den Fokus des Projekts stärker auf die Analyse von ON-Target und OFF-Target Effekten von Kinase-blockern sowie auf molekulare Interaktionen zwischen SARS-CoV-2 und Wirtsenzymen. Um die Spezifität von Wirkstoffen besser zu verstehen, erweiterten wir unsere Enzymaktivitäts-Reporterplattform, um neue Kinasen und das virale MPro zu analysieren. Zusätzlich etablierten wir weitere Read-outs zur Untersuchung von MPro-Aktivitäten und Resistenzen. Diese Assays ermöglichten es uns, die Spezifität und Wirksamkeit gut charakterisierter sowie weniger gut validierter Wirkstoffe zu analysieren. Parallel legten wir einen starken Fokus auf OFF-Target-Effekte von Wirkstoffen auf mitochondriale Funktionen, Phosphorylierungsdynamiken und dem Metabolom. In diesem Zusammenhang können wir nun auch über PINK/Parkin-Reporter interne und externe Auslöser der Mitophagie identifizieren. Ein weiterer wichtiger Schritt des MUI-Teams war die Validierung von SARS-CoV-2-Infektionsassays. Dabei wurde ein optimierter VSV-basierter Pseudovirus-Assay etabliert, der eine zuvor beobachtete Diskrepanz zwischen authentischem Virus und Pseudovirus-Readouts auflöste. Insgesamt schuf das Projekt eine vielseitige Plattform zur Untersuchung von ON-OFF Wirkstoffeffekten auf Wirts- und virale Enzymfunktionen, mitochondriale Aktivität sowie die SARS-CoV-2-Biologie.
