Der Wnt-Signalweg spielt eine wichtige Rolle während der Embryonalentwicklung. Eine strenge Kontrolle des Wnt-Signalwegs ist unerlässlich und erfolgt weitgehend durch die Regulierung der freien Mengen des Proteins ß- Catenin durch den ß-Catenin Destruction Complex. Störungen in der Regulation des Wnt Signalwegs sind eine Hauptursache für eine Vielzahl von Erkrankungen wie Krebs. Jedoch sind die zugrundeliegenden molekularen Mechanismen größtenteils unbekannt. Dies liegt primär an der intrinsischen Flexibilität der involvierten Proteinkomplexe was deren strukturelle Untersuchung mittels Standardtechniken der Strukturbiologie erschwert. Hier schlagen wir vor eine innovative Kombination struktureller Techniken in Lösung zu verwenden, um die molekularen Mechanismen des ß-Catenin-Fangens und Phosphorylierens anhand eines minimalen Axin-1 - GSK3ß - ß-Catenin Destruction Complex zu untersuchen. Dieser minimale Komplex enthält alle, für das ß- Catenin-Fangen und Phosphorylieren benötigten, Teile. Wir sind bestrebt, Struktur und Funktion des Axin-1 GSK3ß - ß-Catenin Komplexes aufzuklären, dessen (Fehl-)Regulation durch post-translationale Modifikationen (PTMs, i.d.R. Phosphorylierung) und krankheitsrelevante Mutationen, sowie funktionellen Implikationen in zellbasierten Assays. Um die, durch die intrinsische Flexibilität des Proteinkomplexes auferlegten Herausforderungen, zu überwinden, werden wir unsere jüngsten methodischen Leistungen für die Untersuchung der Struktur und Dynamik von Proteinen und Proteinkomplexen ausnützen. Dazu verwenden wir einen multidisziplinären Ansatz der Strukturbiologie, in dem wir Lösungstechniken wie Kernresonanz (NMR)- Spektroskopie, Kleinwinkel Röntgen-/Neutronenstreuung (SAXS/SANS) und Molekulare Modellierung kombinieren. Unsere strukturellen und funktionellen Untersuchungen des dynamischen Axin-1 - GSK3 - ß- Catenin-Komplexes werden nicht nur biologisch hoch relevante Einblicke in die allosterische Regulation des ß- Catenin Destruction Complex und Krankheitsmechanismen liefern, sondern auch die Schlüsselrolle ungeordneter Proteine in allosterischer Regulation aufzeigen.
